I think quite a bit more information is needed here. Reason I'm chipping in as I sometimes have a similar problem that may assist here.

Occasionally I have a failure on central locking. Remote lock/unlock don't trigger the door locks. I cannot therefore use the power doors on either side, cannot open them manually (as they're locked!) and so have to resort to using the key to lock drive and passenger doors and struggle.
To solve it, I take the positive battery lead off for about a minute, put it back and it generally kicks back into life. It's like an electronic trip somewhere goes, but its not a fuse, more like an e-fuse.

Have you tried taking the battery lead off for a minute or two and reconnecting, try the central locks and see if it comes back on?

Otherwise, more info:
What model (diesel, petrol, 2.8, 3.5, etc)
Problem itself, is it just the remote keyfob that doesn't work, does the central lock switch in the car work, do the doors lock/unlock.
Is it Left or Right hand drive. Reason I say that is if its right hand drive (UK like mine), then my rear sliding door and drivers door tend to not be locked when I have the fault!
